Hoodia Authenticacy Test Methods

Hoodia Authenticacy Test MethodThree tools that can be used to verify the presence and purity of Hoodia Gordonii in Hoodia products are  examination under a microscope, High Performance Thin Layer Chromatography, and High Performance Liquid Chromatography.  These methods complement one another and should be used together to determine the identity and purity of a raw powdered sample.


A qualified microscopist who has been trained to analyze botanical powders can detect cell components that indicate the presence of what is being looked for, and also negative markers that would not be found in the substance being tested for.  Microscopic features that only occur in Hoodia Gordonii have not yet been found.  So microscopy cannot differentiate with certainty between Hoodia Gordonii and other Hoodia species, or another plant species called Caralluma fimbriata.  But the presence of certain cell types and cellular substructures, or the presence of calcium oxalate crystals, would mean that something in the sample was not from any Hoodia species.

Microscopy cannot positively identify Hoodia Gordonii all by itself.  But it can detect added impurities like Opuntia or maltodextrin even at small concentrations.

High Performance Thin Layer Chromatography

High Performance Thin Layer Chromatography, or HPTLC, gives a visual display of the chemical compounds present in a test sample.  It cannot always rule out the presence of related species or every possible impurity.

The HPTLC technique is a very good tool for identifying Hoodia Gordonii by the characteristic image associated with it.  But more work needs to be done to find the variability range for a genuine sample and to determine how good a tool it is for finding impurites.

High Performance Liquid Chromatography

High Performance Liquid Chromatography, or HPLC, has had recent advances that make it a very good tool for finding specific target molecules in a complicated sample like an herbal dietary supplement.  How well it works depends on the skill of the analyst doing the work, which HPLC method is used, information references used, and the nature of the sample itself.  The two main HPLC methods used are to look for a particular marker, or using a profiling or “fingerprinting” technique.

Which method is used can depend on the ease of doing the analysis, marketing reasons, or other reasons.  When a method is selected it must be based on the question being answered.  The question to answer in this case is just, “Is this Hoodia Gordonii or isn’t it?”

However, this is actually the hardest type of question to answer in a laboratory analysis.  For a definite answer, determining features that are either chemical, genetic, or structural must be used to tell Hoodia Gordonii apart from other Hoodia species and impurities known to be used.  The famous P57 molecule and other compounds related to it are used to determine that a Hoodia species is present.  P57 itself is considered a marker for Hoodia Gordonii, but this is not necessarily the end of the story, because some P57 can be found in other Hoodia species.  And the image generated by P57 can coincide with elements of the visual displays generated by some impurities, for instance, Gymnema sylverstre.  There is a way now to increase the HPLC run time so that the images for Gymnema sylvestre and P57 split out from one another.  This improved method clearly shows 11 different P57-related compounds characteristic of Hoodia Gordonii.  More work is being done to create a better chemical portrait of Hoodia Gordonii and to differentiate it from more impurities.
